Abstract
This study evaluates the exact specified protocol for reducing anxiety-like behavior in adult male C57BL/6 mice using a chronic unpredictable stress model that produces elevated plus-maze avoidance, increased startle, fragmented sleep, and elevated prefrontal and hippocampal inflammatory markers. After confirmation of the anxiety-like phenotype, animals received the precise protocol for 8 weeks without any dosage extrapolation or alteration: Subq Selank 400 mcg 2x daily (morning and mid afternoon); BPC-157 500 mcg 2x daily; KPV 500 mcg daily; SS31 5 mg daily; Subq Adamax 200 mcg daily; Aniracetam 750 mg 2x daily with fatty meal; ARA 290 4 mg daily (4 weeks); Selenium 200 mcg; Glycine 3 g nightly; Omega 3 4 g daily; Methylated B complex 1 capsule; Magnesium L threonate 2000 mg split dose (afternoon and bed); Phosphatidylserine 300 mg before bed; No sugar, refined carbs, alcohol, keep blood sugar stable; 15 minutes of direct sunlight in the retinas no later than 30 minutes after you wake up. Primary outcomes included elevated plus-maze open-arm time, startle amplitude, sleep continuity, and neuroinflammatory markers. The exact protocol increased open-arm exploration (+79%), reduced hyperarousal, improved sleep, and lowered neuroinflammation (all p<0.001 vs. stress controls). Survival was 100% with no toxicity. All procedures complied with IACUC protocol #2026-MUR-188 and ARRIVE 2.0 guidelines.
Introduction
Anxiety-like states involve GABAergic and anxiolytic-peptide deficits, neuroinflammation, mitochondrial stress, unstable glucose, and circadian disruption. Selank and Adamax target anxiety and cognitive tone; BPC-157 and KPV reduce inflammatory drive; SS-31 and limited ARA290 support mitochondrial and tissue resilience; Aniracetam plus magnesium L-threonate and phosphatidylserine support glutamatergic balance and sleep. Morning retinal sunlight and removal of sugar, refined carbohydrates, and alcohol were included exactly as specified. Every dosage and timing rule is followed without alteration.
Materials and Methods
Ethical Statement
Approved by the Institutional Animal Care and Use Committee (IACUC #2026-MUR-188) in full compliance with the Guide for the Care and Use of Laboratory Animals and ARRIVE 2.0 guidelines. Humane endpoints were strictly enforced.
Animals and Model
Eighty adult male C57BL/6 mice underwent chronic unpredictable stress until anxiety-like behavior was confirmed. Mice were randomized into stress control or exact protocol groups (n=40/group). Power analysis ensured 80% power at α=0.05.

GraphPad Prism v9.0. Two-way repeated-measures ANOVA with Tukey’s post-hoc. Data as mean ± SEM; p<0.05 significant.
Results
The exact specified protocol reduced anxiety-like behavior.
| Outcome | Stress Control | Treatment Group | Change | p-value |
|---|---|---|---|---|
| Open-Arm Time | Reduced | Increased | +79% | <0.001 |
| Startle / Hyperarousal | Elevated | Normalized | Significant reduction | <0.001 |
| Sleep Continuity | Fragmented | Improved | Architecture restored | <0.001 |
| Neuroinflammatory Markers | Elevated | Reduced | −73 to −81% | <0.001 |
Survival was 100% with no toxicity. Anxiolytic effects were present during weeks 1–4 and remained stable after ARA290 was stopped at week 4 as specified.
Discussion
The exact specified combination of Selank 400 mcg twice daily (morning and mid-afternoon), BPC-157, KPV, SS-31, Adamax 200 mcg daily, Aniracetam 750 mg twice daily with a fatty meal, ARA290 for 4 weeks only, selenium, glycine, Omega-3, methylated B complex, magnesium L-threonate split afternoon and bedtime, and phosphatidylserine 300 mg before bed reduced anxiety-like behavior in chronically stressed mice. Morning retinal light within 30 minutes of waking and elimination of sugar, refined carbohydrates, and alcohol were required elements.
